Ninhydrin-positive Substances by HPLC using Post-Column Derivatisation

Butterworth Labs is one of the first laboratories in Europe to comply with the latest European Pharmacopoeia (Ph Eur) monographs of 17 amino acids, analysing ninhydrin-positive substances by High Performance Liquid Chromatography (HPLC) using post-column derivatisation with ninhydrin and dual wavelength detection as per method 1 in the general monograph 2.2.56.

The system uses a gradient with two buffers and a pre-prepared Biochrom EZ Nin reagent.

The recent changes to the ninhydrin-positive substances Ph Eur test method affect the following monographs:

Alanine

Arginine

Arginine hydrochloride

Cysteine hydrochloride monohydrate

Glycine

Histidine

Histidine hydrochloride monohydrate

Isoleucine

Leucine

Lysine hydrochloride

Phenylalanine

Proline

Serine

Threonine

Valine
